Moderate Osmotic Stress Ameliorates Root Formation and Acclimatization in Apple Rootstock

Apples are regarded as one of the most significant temperate fruits. However, their rooting process is often challenging due to low auxin levels. This study aimed to explore the impact of various stress conditions on the rooting efficiency of apple rootstock (MM111). For this purpose, the microshoot explants were exposed to osmotic (1.5X and 2X sucrose), cold (18 °C), heat (28 °C), and wounding stress conditions, and factors related to rooting were recorded at the end of the experiment. The findings of this study revealed that explants exposed to osmotic stress and heat stress exhibited the highest root formation percentage and the greatest number of roots. Wound stress caused a significant increasing in browning of explants. Also, control treatment and moderate osmotic stress recorded the highest success in acclimatization. In conclusion, controlled exposure of explants to stress conditions, especially mild osmotic stress, stimulated the microshoots to induce rooting. Also, this treatment plays a key role in improving the survival of explants during the acclimatization phase.
